What Does HPLC Tell Us About Research Peptides?
HPLC is one of the most common testing methods shown on a research peptide Certificate of Analysis. The name may sound technical, but its purpose is fairly simple: it helps researchers look at what is present in a sample and estimate its purity.
WHAT DOES HPLC MEAN?
During testing, a small sample is passed through specialized equipment that separates its different components. The results are displayed in a graph called a chromatogram.
This allows the laboratory to see the main component as well as smaller amounts of other material detected in the sample.
HOW DOES HPLC WORK?
Think of HPLC as a sorting process.
The different components in a sample move through the testing equipment at different speeds. Because they do not all reach the detector at the same time, the equipment can separate them and display each one as a peak on the graph.
The result gives the laboratory a closer look at what makes up the sample.
WHAT DO THE PEAKS MEAN?
An HPLC graph usually contains one large peak and several smaller ones.
You do not need to study every line on the graph. The most important things to look for are:
WHERE DOES THE PURITY PERCENTAGE COME FROM?
The purity percentage is calculated by comparing the main peak with the other peaks detected during the test.
For example, if a report shows 99% purity, it generally means that approximately 99% of the material detected by that method was associated with the main component.
This can provide useful information about the sample, but the number should always be considered alongside the rest of the COA.
WHAT HPLC DOESN’T TELL US
HPLC provides helpful information about purity, but it does not answer every question about a research peptide.
A purity result does not automatically confirm the peptide’s identity, the amount in the vial, or whether other types of testing were performed. Those details require separate testing and should be clearly listed on the COA.
WHY HPLC AND MASS SPECTROMETRY ARE OFTEN USED TOGETHER
HPLC and mass spectrometry look at different parts of a sample.
HPLC helps researchers examine purity, while mass spectrometry helps confirm identity. When both results are available, they provide a more complete look at the research peptide than either test could provide alone.
MAKING SENSE OF THE RESULTS
You do not need to interpret every line or peak to learn something from an HPLC report.
Start with the reported purity, check that the results match the correct batch, and confirm that the supporting graph is included. Knowing these basics makes HPLC results much easier to follow.
